A. Standard Methods Media
(a) Dilution water.To prepare stock solution, dissolve 34 g KH2PO4 in 500 mL H2O, adjust to pH 7.2 with 1M NaOH (ca 175 mL), and dilute to 1 L withH2O. To prepare bufferedH2Ofor dilutions, dilute 1.25 mL stock solution to 1 L with boiled and cooled H2O. Autoclave 15 min at 121°C.
(b) Buffered glucose broth (MR-VP medium).For methyl red-Voges Proskauer (MR-VP) tests. Dissolve 7.0 g proteose peptone, 5.0 g glucose, and 5.0 gK2HPO4 in ca 800mLH2Owith gentle heat and occasional stirring. Filter, cool to 20°C, and dilute to 1 L. Dispense 10mLportions into test tubes and autoclave 1215 min at 121°C. Maximum exposure to heat should be £30 min. Final pH, 6.9 ± 0.2.
(c) Endo medium.Suspend 3.5 g K2HPO4, 10.0 g peptone, 20.0 g agar, and 10 g lactose in1LH2O. Boil to dissolve, addH2O to original volume, and clarify if necessary. Dispense in 100 mL portions and autoclave 15 min at 121°C. Final pH, 7.4 ± 0.1. Before use, melt and add 0.25 g Na2SO3 and 1.0 mL filtered 5% alcohol solution basic fuchsin.
